Abstract

Gljiva Sphaeropsis sapinea je patogen iglica, kore i drva različitih vrsta borova. Prilikom izolacije micelija sa crnog bora (Pinus nigra), korištene su dvije različite metode, metoda kotrljanja plodišta te metoda nanošenja suspenzije spora. Dobiveni miceliji značajno su se razlikovali u izgledu i broju micelija. Izgled dobivenih micelija promatran je na dvije hranjive podloge, Malt Extract Agar (MEA) i Potato Dextrose Agar (PDA). U istraživanju je dokazano da gljiva na obje hranjive podloge postiže zadovoljavajući rast i razlike u izgledu nisu uočene. Dobiveni miceliji gljive dugoročno su pohranjeni na dvije metode, prvom metodom u 15 % glicerol na -80 °C, a drugom metodom komadići micelija su pohranjeni u 50 % glicerol na 4 °C.

Abstract (english)

Sphaeropsis sapinea fungus is a pathogen of different species of pine trees, their bark and needles. During the mycelium isolation from the black pine (Pinus nigra), two different methodes were used, method of rolling the fruiting body and spores solution applying method. The resulting mycelia have segnificantly differed in the shape and number of mycelium. The shape of resulting mycelia was opserved on two types of agar, Malt Extract Agar (MEA) and Potato Dextrose Agar (PDA). The result of experiment prove that the fungus reaches satisfactory growth on both agars and differences in shape or form have not been observed. Resulting fungus mycelia have long term been stored in two ways, first method was 15% glycerol at – 80 °C, and the second method, pieces of mycelia were stored in 50 % glycerol at 4 °C.
