Abstract

U ovom radu uzorkovano je 156 rameta 52 klona hrasta kitnjaka iz klonske sjemenske plantaže „Novoselci“. Klonska sjemenska plantaža osnovana je pomoću vegetativnih kopija izabranih majčinskih stabala. Kopije su proizvedene cijepljenjem. Jedinke su analizirane pomoću 14 jezgrinih mikrosatelitskih biljega kako bi se odredili genotipovi svih klonova. Uspješno su se amplificirali odsječci DNA za 155 jedinki, od kojih je 130 pridruženo nekom klonu. 20% jedinki nije odgovaralo nacrtu. Neke su krivo obilježene, a kod nekih je došlo do odumiranja plemke i biljka se razvila iz podloge. Uzorci su ispravljeni u nacrtu plantaže. Jedinke koje su nastale rastom podloge preporuča se ukloniti zbog negativnog utjecaja n agenetsku dobit. Genetska raznolikost genotipova klonova zadovoljavajuća je i odgovara vrijednostima iz prirodnih sastojina hrasta kitnjaka, te predstavlja dobru genetsku bazu za proizvodnju sjemena (HE = 0,817).

Abstract (english)

In this study, 156 ramets from 52 clones of sessile oak were sampled from the "Novoselci" clonal seed orchard. The clonal seed orchard was established using vegetative copies of selected parent trees. The copies were produced through grafting. The individuals were analysed using 14 nuclear microsatellite markers to determine the genotypes of all clones. DNA fragments were successfully amplified for 155 individuals, of which 130 were assigned to a clone. Twenty percent of the individuals did not match the layout plan. Some were mislabelled, and in some cases, the scion had died and the plant developed from the rootstock. The samples were corrected in the orchard layout. It is recommended to remove individuals that developed from the rootstock due to their negative impact on genetic gain. The genetic diversity of the clone genotypes is satisfactory and corresponds to the values found in natural sessile oak stands, representing a good genetic base for seed production (HE = 0.817).
