Abstract

U klonskoj sjemenskoj plantaži u Kutini prikupljeni su izbojci,za potrebe mikropropagacije, uzeti s 9 različitih klonova. Sa svakog klona (stabla) prikupljena po 4 izbojka s pupovima u fazi dormancij; po jedan izbojak na vrhu i na dnu krošnje sa sjeverne strane svijeta te po jedan izbojak na vrhu i na dnu krošnje sa južne strane svijeta. Prikupljeni izbojci poslužili su kao izvor eksplantata za uvođenje u kulturu tkiva metodom meristema. Sa svakog prikupljenog izbojka u kulturu tkiva je uvedeno po 6 pupova, što ukupno po klonu čini 24 uzorka. Inokulirane kulture svoj su razvoj nastavile u klima komori u razdoblju od 40 dana prilikom čega je praćen njihov razvoj. Cilj rada bio je utvrditi uspješnost i brzinu napretka razvoja biljčica pojedinog klona u kulturi tkiva, s obzirom na poziciju uzetog uzorka u krošnji, kao razlike u razvoju i preživljenju klonova međusobno, zbog utjecaja genetske raznolikosti.

Abstract (english)

In the clone seed plantation in Kutina, shoots were collected, for micropropagation purposes, taken from 9 different clones. From each clone (tree) were collected 4 shoots with buds in the dormancy phase; one shoot at the top and bottom of the canopy on the north side of the world and one shoot at the top and bottom of the canopy on the south side of the world. The collected shoots served as a source of explants for introduction into tissue culture by the meristem method. From each collected shoot, 6 buds were introduced into the tissue culture, which makes a total of 24 samples per clone. Inoculated cultures continued their development in the climate chamber for a period of 40 days, during which their development was monitored. The aim of this study was to determine the success and speed of progress in the development of individual clone plants in tissue culture, given the position of the sample taken in the canopy, as differences in the development and survival of clones, due to the influence of genetic diversity.
